چکیده

Statistical experimental design was employed to optimize medium compositions to enhance the activity of lipase from Bacillus subtilis. Plackett–Burman design was applied to evaluate the effect of different medium compositions obtained by one-factor-at-a-time experiments, and ammonium chloride, ammonium sulfate and dipotassium hydrogen phosphate were found to influence the activity of lipase significantly. Steepest ascent method was employed to approach the experimental design space, followed by an application of central composite design and response surface methodology for further optimization. A quadratic model was found to fit the lipase activity, and the optimal medium compositions were determined as included 45 g L NH4Cl, 20.4 g L –1 (NH4)2SO4 and 12.6 g L K2HPO4. The corresponding lipase activity was 91.39 U mL . Validation experiments were also carried out to prove the adequacy and the accuracy of the model obtained. The lipase activity reached 91.46 U mL, which was almost equal to the actual predicted value, about 4.94-fold increase compared with that using the original medium.

Optimization of Lipase Production Using Bacillus subtilis by Response Surface Methodology

A total of 6 isolates of Bacillus subtilis were isolated from oil mill waste collected in Namakkal district, Tamilnadu, India. The isolated bacteria were screened using lipase screening medium containing Tween 80. BS-3 isolate exhibited a greater clear zone than the others, indicating higher lipase activity. Therefore, this isolate was selected for media optimization studies. Ten process variab...

Secretion of cyclodextrin glucanotransferase in E. coli using Bacillus subtilis lipase signal peptide and optimization of culture medium.

The cyclodextrin glycosyltransferase (CGTase) of Paenibacillus pabuli US132 was fused to the secretive lipase signal peptide of B. subtilis. This leads to an efficient secretion of the recombinant enzyme into the culture medium of E. coli as an active and soluble form contrasting with the native construction leading to a periplasmic production. In order to enhance the yield of CGTase production...

Molecular Detection of Lipase A gene in Putative Bacillus subtilis Strains Isolated from Soil

The present study was undertaken to screen the soil samples collected in Iran for the presence of the Bacillus subtilis lipase A gene. The bacterial colonies obtained from the collected soil samples were examined by physical appearance, biochemical tests and  the polymerase chain reaction (PCR). Only four colonies were identified as putative B. subtilis strains and all contained the lipase A ge...
